Comparative RNA-Seq transcriptomic analyses of ten ecotypes of Phaeodactylum tricornutum: toward identifying the best cell biofactory for the production of biologics

Résumé

Most biologics are glycoproteins produced in mammalian cells. However, this industrial process is associated with some limitations such as high production costs, mainly due to the long purification processes of the bioproducts. Nevertheless, microalgae emerged as a cheaper alternative cell biofactories, due to their lack of common pathogens with humans, their rapid growth rate, and their ability to perform post-translational modification of proteins. Among them, the diatom Phaeodactylum tricornutum has numerous advantages and has been successfully used to produce biologics such as SARS-COV2 RBD1 and functional monoclonal antibodies (mAbs)2-4. These mAbs have been shown to be glycosylated with mammalian-like oligomannosides that result from processing steps occurring in the ER and the early Golgi apparatus3. It is well established that N-glycans associated to proteins are involved in the half-life, bioactivity and immunogenicity of proteins. Currently, despite the successful production of biologics in P. tricornutum, the yields obtained are actually too low to be competitive at an industrial scale. Recently, the genomes of ten ecotypes of P. tricornutum have been sequenced. It is therefore essential to better understand the cell biology of the ten ecotypes of P. tricornutum and unravel criteria for the selection of the best P. tricornutum strain for the bioproduction of biologics. In the present work, we performed RNA-Seq analyses on the ten ecotypes of P. tricornutum and decode the differential expression of genes involved in several pathway that could have an impact on the production of biologics products such as production yields, secretion or quality of the bioproducts.
